いよいよ大詰めね。覚悟なさい。

The character is talking to herself (since there is no one else around), but it seems strange to me that she would use なさい to herself. I see something like "get ready" but I can't work out the details. I must get ready? Is she telling the enemy to prepare? :?

I believe she's telling the enemy to get ready (for getting beaten up) as you say.

It's an old school shoot em up called 銀河婦警伝説. Very little text in the game, a good way to start learning. My character will say a single sentence or two at the end of each level in response to the enemy escaping (which he does at the end of every level).

in response to the enemy escaping

are two very different contexts. Imagine watching a movie where two characters are arguing, and then one storms out of the room. The other then says/yells something along these lines:

"Jerk."
"And don't come back!"
"This isn't over!"

etc.etc.

Even though the other person isn't in the room and may not be able to hear them, would you really say this speaker is "talking to themselves"? It's the context that makes it a final comment at a fleeing enemy, not a person standing in an empty room who suddenly starts yelling "I'LL GET YOU NEXT TIME!" at empty air.
