How are you suppose to reply to はじめまして

You can reply back with はじめまして...it basically translates to "nice to meet you" and is used in a fashion similar to English. Since the person is introducing themselves to you, it would be rude of you to not do the same in return.
 
But I thought はじめまして was "How do you do?"

And what's the difference between はじめまして and どうぞ宜しく?

Not really. The nearest to "How do you do?" is お元気ですか。"
 
There are several possiblities, but something alone these lines should do.

Hajimemashite. (Your name desu.) (Kochirakoso.) Douzo yoroshiku (onegaishimasu.)
