欠員 is vacant position/post, i.e., for people, not for time.

What's the most suitable word for general worker in restaurant or convenience store?

In what meaning are you using "time", time period of a day or season?
Th initial one いつですか is correct for a question regarding time.

(person人/worker 員 ) はいつ必要ですか？
What's the most suitable word for general worker in restaurant or convenience store?

店員 or 従業員 is the word, but I would use other expressions where that word is not mentioned, for instance, 人手が必要なのはいつ頃ですか.
