it means persistent, unrelenting, possibly to the point of being of annoying. The connotation usually isn't positive.

Honestly, if she is calling you shitsukoi multiple times, you probably shouldn't reply at all.
Whatever you're doing (even if it's just talking to her) is obviously bothering her, and she clearly just wants to be left alone.

Anyone can say it, though it definitely is a common expression for women to use when men are making unwelcome romantic advances toward them.
