If I use 日本の主要な島 does that normally mean the 4 largest islands?
I would like to say that next years vacation will be spent touring japans 4 main islands i.e excluding 沖縄、by train.
Would something like this be okay?
来年お休みは日本の主要な島中に電車で旅行します
Or would you have to stipulate the four largest islands? not sure how I would say that possibly 日本の上位４最大の島?

The particle for the location you travel about is を, btw. に is for the location you travel to, i.e., 日本を旅行する and 日本に旅行する have different meanings.
